摘要
目的研究血小板衍生内皮细胞生长因子(PD-ECGF)及Pan caspase-1在胃腺癌组织中的表达,进一步探讨PD-ECGF在胃腺癌血管形成中的作用及Pan caspase-1对胃腺癌细胞凋亡基因蛋白的影响,为临床正确判断肿瘤细胞的生物学行为及估计患者预后、筛选可靠生物学指标。方法收集2001-2003年本院胃腺癌手术标本蜡块54例。其中低分化胃腺癌32例,高分化22例。采用SP免疫组化法,严格按照SP超敏试剂盒的说明操作,显微镜下观察PD-ECGF和Pan caspase-1在胞浆/胞核中的表达。并结合F-ⅧR Ag检测,测量MVD值。结果PD-ECGF在低分化腺癌表达为75.0%,高分化表达为36.4%,两者有显著性差异(P<0.05)。Pan caspase-1在低分化腺癌表达为21.9%,高分化表达为72.8%,两者有显著性差异(P<0.01)。肿瘤的MVD在PD-ECGF和Pan caspase-1的阳性表达中分别高于其阴性表达(P值分别是0.0033和0.0149)。结论PD-ECGF有助于胃腺癌间质血管形成,与癌组织分化程度有关。Pan caspase-1在癌细胞凋亡有作用,与癌组织分化程度有关。提示联合检测PD-ECGF和Pan caspase-1有助于确定癌细胞的分化程度,正确判断胃癌细胞的生物学行为及患者预后。
Objective To investigate the relationship between tumor angiogenesis and the expression of platelet - derived endothelial cell growth factor/thymidine phosphorylase (PD - ECGF/dThdPase). Although it is reported that Pan caspase - 1 protein was involved in apoptosis, apoptosis effects mutually generated MVD and Pan caspase- 1 have not yet been evaluated. Methods The expressions of PD- ECGF and Pan caspase - 1 in 54 gastric adenocarcinoma tissue samples, including 32 low differentiated gastric adenocarcinoma and 22 well differentiated gastric adenocarcinoma were assessed immunohistochemically, and the microvessel density (MVD) was determined by immunostaining for factor VIII- related antigen. Resuits Out of the 32 low differentiated gastric adenocarcinoma tissue samples, 24 (75,0 % ) were PD - ECGF positive, and 7 (21. 9%) were Pan caspase- 1 -positive; out of the 22 well differentiated gastric adenocarcinoma tissue samples, 8 (36.4 % ) were PD- ECGF - positive, 16 (72.8 % ) were evaluated as Pan caspase- 1 - positive. The expressions of PD-ECGF and Pan caspase - 1 were significantly associated with degrees of differentiated gastric carcinoma ( P 〈 0.05, P 〈 0.01 ). The microvessel density was significantly higher in PD- ECGF- and Pan caspase- 1 - positive tumors than that in PD - ECGF - and Pan caspase - 1 - negative tumors ( P = 0. 033, 0.0149). Conclusions The results suggest that expression of PD - ECGF plays a role in the promotion of angiogenesis in human gastric adenocarcinoma and MVD has effect on expression of Pan caspase- 1, which induces apoptosis in human gastric adenocarcinoma.
