摘要
目的:探讨目标化合物γ-(N-对甲基苯磺氨基)丙基-3,7,10-三甲基-2,8,9-三氧杂-5-氮杂-1-硅杂三环[3,3,3,01,5]十一碳烷对人宫颈癌细胞(Hela)、人结肠癌细胞(SW480)和人肺腺癌细胞(A549)的抗肿瘤效应以及对单子叶植物玉米、双子叶植物萝卜的生长调节作用。方法:将目标化合物用培养液配制成浓度分别为20μg·ml-1、100μg·ml-1和200μg·ml-1的溶液,采用四甲基偶氮唑盐比色分析法(MTT法)测试目标化合物对Hela细胞、SW480细胞和A549细胞的体外抗肿瘤活性。将目标化合物用水配制成浓度分别为10ppm、50ppm和100ppm的溶液,采用平皿法测试目标化合物对单子叶植物玉米和双子叶植物萝卜的生长调节作用。结果:目标化合物对三种肿瘤细胞株的增殖均具有抑制作用,在测定浓度范围内,随着浓度的增加,抑制率也在增加,呈现良好的剂量依赖性,三种细胞株中对SW480抑制率最高。目标化合物在三种浓度下均有促进玉米和萝卜生长的作用,在50ppm浓度下促生长的效果最好。结论:目标化合物具有体外抗肿瘤活性及促进植物生长的活性。
Objective: To sexplore the anti-tumor activity to Hela, SW480 and A549 and the growth regulative activitiy to radishes of amphibrya besides corns of dicotyledon of the target compound γ - (N-p-methylphenylsttlfonyl)aminopropy1-3,7,10-trimethy1-2,8,9-trioxa- 5 -aza 1 - 1 -silicontricyclo [3,3,3,0^1,5] Undecanes Methods: The concentration of the compound was prepared as 20, 100, 200 μg·ml^-1 using culture media. The anti-tumor activity to Hela, SW480 and A549 was analyzed with MTT method. The concentration of the compound was prepared as 10, 50, 100 ppm using water. Plating method test was used to exmtine its growth regulative activity on radishes of amphibrya and corns of dicotyledon. Results: The target compound showed good inhibition ratio in vitro and the inhibition ratio depended on the cconcentration. The inhibition ratio to SW480 was the highest. The result of the plating method test showed that it could promote plants to grow well and the effect was best under the 50 ppm concentration. Conclusion: The target compound shows antitumor activity and can promote plants growth.
