大鼠海马中83KD蛋白诱导神经干细胞向AChE阳性神经元定向分化的作用 被引量:1

摘要 目的:探讨海马组织中83KD蛋白诱导神经干细胞向ACHE阳性神经元定向分化的作用。方法:将切割海马伞后14天海马和正常海马组织匀浆进行非变性聚丙烯酰胺凝胶电泳,根据染色脱色结果取83KD差异蛋白条带进行电洗脱,将收集到的蛋白质加入到鼠胚神经干细胞球培养液中,并设阴性对照组。12天后,用AChE组织化学染色检测神经干细胞分化为AChE阳性神经元的情况。结果:83KD切割组中AChE阳性神经元较多,胞体较大且分化较好,突起较粗且长;83KD正常组中AChE阳性神经元数量较少,胞体较小,突起也较短;空白对照组仅看到少量的AChE阳性神经元,胞体很小,突起短而少。结论:海马中83KD蛋白具有诱导神经干细胞向AChE阳性神经元定向分化的作用。 Objective: To explore the effect of 83KD Protein in Hippocampi on? Inducing neural stem cells differentiating into AChE positive neurons. Methods: The normal hippocampi and the ones on the 14th day after the fimbria transection were used for 10% Native-PAGE, and the differential protein of 83KD were electroeluted, neural stem cells clone spheres of rats were cultured in 3 groups as follow: 83KD normal group, 83KD transection group and the control group. On the 12th day after culture, Acetylcholinesterase histochemical stain was used to detecte the differentiation of neural stem cells into cholinergic neurons. The number, dimensions and perimeters of AChE positive neurons were analyzed with image analysis system. Re- sults: The AChE positive neurons in 83KD transection group were the most and its bodies were very big and the processes were abundant; The AChE positive neurons in 83KD normal group were inferior to those in 83 transection group, There were few AChE positive neurons in control group, Conclusion: 83KD protein can induce neural stem ceils differentiating into AChE positive neurons.
