摘要
目的了解深圳市2007年H3N2亚型流感病毒流行情况及血凝素基因特性。方法用狗肾传代细胞(MDCK)和鸡胚进行流感病毒分离。收获病毒后提取病毒RNA,进行逆转录-聚合酶链式反应(RT-PCR),扩增产物经琼脂糖凝胶电泳鉴定后送大连宝生物公司纯化及测序。测序结果用Simmonic软件和Mega3.1软件进行序列分析。结果H3N2亚型流感毒株为深圳市2007年流感流行的优势株,与2006年的深圳株比较未发生明显变异,但与2007年的疫苗株相比,有多个位点的氨基酸发生了替换,造成了其间的抗原性发生了一定的变化。结论深圳市2007年H3N2亚型流行株,并未形成一个新的变种。推测是由于与疫苗株之间的抗原性的变异导致了2007年深圳市H3N2亚型流感的流行。
Objective To understand the prevalence and genetic characterizations of group A influenza virus H3N2 in Shenzhen in 2007. Methods Influenza viruses isolated were cultured in MDCK cell and embryonated eggs. Virion RNAs were extracted and amplified by using RT - PCR. The amplified products were sent to Dalianbao Biological Company for purification and sequencing after identified by gel electricity . The results were analyzed by SIMMONIC and MEGA3.1 software. Results H3N2 subtype of influenza strain was dominant in Shenzhen in 2007. There no significant variation was observed compared to that of 2006 ; while replacements of amino acids was observed at multiple sites by comparison with the vaccine strain in 2007, which resulted in certain antigenic changes. Conclusion The epidemic strain of H3N2 subtype is not a new variant in shenzhen in 2007. The H3N2 subtype influenza maybe is caused by the variations of the antigenicity between vaccine strains.
