摘要
目的研究神经元特异性烯醇酶(Neuron-specific enolase,NSE)的改变,探讨液化石油气(LPG)中毒后脑组织的损坏情况。方法采用大鼠液化石油气中毒模型,动物分别于中毒后1、3、7、14、21、28d处死,苏木精-伊红染色观察脑组织神经细胞的动态变化,免疫组化方法检测脑组织NSE的表达水平。结果大鼠液化石油气中毒后大脑皮层神经元细胞核浓缩,结构不清,随中毒浓度增加,细胞核浓缩更明显,可见神经元坏死迹象。免疫组织化学技术观察显示NSE表达增加,随中毒浓度升高,NSE表达亦明显增加。结论液化石油气中毒后大脑皮层神经元形态学改变是认知功能下降的病理基础之一,脑组织NSE表达的增加是脑组织损伤的一种表现。
Objective To evaluate the effect of liquid petroleum gas (LPG) upon brain damage and the expression level of neuron - specific enolase (NSE) in rats after acute liquid petroleum gas poisoning. Methods Liquid petroleum gas poisoning model rats was established. 1,3,7,14,21 and 28 days after poisoning, the rats were killed respectively, the morphological changes in neuron were observed by HE staining, the expression levels of NSE in brain was detected by using immunohistochemistry. Results The degeneration of neurons and inspissation of karyon were observed in LPG treated rats; and the expression of NSE was increased along with the increase of treatment with LPG. Conclusion The changes in morphology of neurocyte after acute liquid petroleum gas poisoning is one of the mechanisms of cognitive dysfunction. The increased expression of NSE in brain is an injury phenomenon.
