墓头回诱导脐血基质细胞生长促粒-单系和红系造血祖细胞集落形成的研究 被引量:1

The effect of ixeritis herba induced stromal cells proliferation and contributed colony formation of CFU-GM and CFU-E progenitor in umbilical cord blood

摘要 目的研究中药墓头回对脐血基质细胞促粒-单系和红系造血祖细胞集落形成的影响。方法收集12例脐血单个核细胞(MNC),加入不同浓度的墓头回水提物共培养14天,基质细胞层形成后,消化,计数;收集基质细胞最多浓度的上清液(MTH-PSCS),以不同比例与MNC共同加入集落培养基中共培养14天,计数粒-单系和红系造血祖细胞集落数。结果0.01μg/ml的墓头回与MNC共孵育14天后,基质细胞的数量(14±2.8)×105/ml明显多于对照组(9±2.2)×105/ml。0.01μg/ml的MTH-PSCS以0、5、10、20比例作用后,脐血粒-单系造血祖细胞集落数分别为17±6、121±24、268±27、114±31、68±11,10%的MTH-PSCS作用最为明显;脐血红系造血祖细胞集落数分别为38±12、72±23、81±9、159±45、90±32,其中以20%的MTH-PSCS作用最为明显。结论中药墓头回可诱导脐血基质细胞的生长,可以促进粒-单系和红系造血祖细胞集落的形成。 Objective To study tile effect of ixefitis herba inducing stromal cells proliferation and enhancing CFU-GM and CFU- E progenitor colony formation in umbilical cord blood. Methods After umbilical cord blood gathered, the blood was separated with mononuclearcells separating medium into umbilical cord blood mononuclearcells. Different concentration of ixeritis herba aqueous extract and the mononuclearcells cohabited for 14 day, then the cells adhered to culture flask was numbered after digestion with trypsin. After gathering supernant of ixeritis herba cohabited with mononuclearcells for 14 day, the supernant that was dilluted to 5,10,20% respertively with contain of mononuclearcells was added to cluny culture media. And 14 days late, CFU-GM and CFU-E progenitor colonys were numbered under microscope. Results Ixeritis herha of 0.01 μg/ml notedly increased the stromal cells number up to (14 ± 2.8) × 10^5/ml, compared to the control" group number of(9 ± 2.2) × 10^5/ml, as others concentration of ixeritis herba not. Cohabited with MTH-PSCS, which was supemant got from Ixeritis herba of 0.01 μg/ml cohabited with nononuclearcells, at the concentration of 5,10,20% for successive 7 days, CFU-GM progenitor colony number was up to 121±24,268 ± 27,114± 31,68 ± 11, whereas the control group is 17 ± 6, and the CFU-E was up to 72 ± 23,81 ± 9,159 ± 45,90 ± 32 but the control group is 38 ± 12, respectively. MTH-PSCS with concentration of 10 % increased CFU-G group number mosdy as 20 % to CFU-E. MTH-PSCS with concentration of 10 % increased CFU-GM group number mostly as 20 % to CFU-E. Colldusion lxeritis herba can induce stromal cells proliferation and enhance CFU-GM and CFU-E progenitor colony formation in umbilical cord blood.
