槲寄生生物碱通过P53途径抑制肝癌细胞株SMMC-7721的研究 被引量:2

摘要 目的:探讨槲寄生生物碱对肝癌细胞株SMMC-7721 P53基因表达的影响。方法:MTT法测定槲寄生生物碱对人肝癌细胞株SMMC-7721增殖的抑制作用;半定量RT-PCR法测定抑癌基因P53 mRNA表达变化。结果:槲寄生生物碱作用后SMMC-7721细胞出现显著生长抑制作用,且呈剂量和时间依赖性。RT-PCR检测SMMC-7721细胞中P53 mRNA表达的相对强度,发现0.100mg/mL的槲寄生生物碱作用于SMMC-7721细胞24小时后,P53 mRNA表达的相对强度增高(P<0.01)。结论:槲寄生生物碱对抑癌基因P53表达的影响可能是其抑制肿瘤生长的分子基础。 Objective :To examine the effects of mistletoe alkali on P53 expression in Human Hepatocareinoma cell SMMC - 7721. Methods:MTF assay was used to evaluate the inhibitory effects of mistletoe alkali on the proliferation of human Hepatocarcinoma cell SMMC- 7721. RT-polymerase chain reaction (RT- PCR) technique was used to evaluate P53 mRNA expression in human Hepatocarcinoma cell SMMC -7721. Results:Mistletoe alkali significantly inhibits the growth of SMMC -7721 cells in a time and dose-dependent manner. After exposure of SMMC -7721 ceils with mistletoe alkali at 0.100mg/mL for 24 hours, the relative intensity of P53 mRNA expression level as determined by RT - PCR increases markedly ( P 〈 0. 01 ) . Conclusion : The stimulation of the expression of anti - oneogene P53 by mistletoe alkali may underlie the molecular basis for it to inhibit tumor growth
