三种固定液对神经纤维冰冻切片固定效果之比较

Comparison of Fixation Effects of Nerve Fiber Frozen Section by Three Fixatives

目的:观察比较10%甲醛、丙酮和Bouin三种固定液对神经纤维的固定效果,为临床做出更好的神经纤维的快速冰冻切片提供实验基础。方法:取健康大鼠双侧坐骨神经,冰冻切片后分别用三种固定液固定,然后做HE染色,观察各自的组织结构及染色情况。结果:10%甲醛固定的神经纤维组织结构较清楚但是组织发生收缩;丙酮固定的神经纤维染色效果较好,组织没有发生收缩;Bouin固定液固定的神经纤维结构和染色都比较好但是组织收缩最明显。结论:三种固定液都可用于固定神经纤维,但丙酮效果最好。

Objective：To compare the fixation effects of nerve fibers by three fixatives, 10% formaldehyde, acetone, and bouin so as to provide experimental proof for making rapid frozen section of nerve fibers clinically. Methods ： Sciatic nerves dissected from healthy rat were subjected to the technique of frozen section and then followed by fixation, and HE staining. The tissue structure of each HE-stained sciatic nerve frozen section was carefully examined. Results ： Sciatic nerve frozen section fixed by 10% formaldehyde exhibits relatively clear tissue structure but undergoes tissue structure shrinkage; frozen section fixed by acetone exhibits good staining effect with no tissue structure shrinkage observed; frozen section fixed by bouin exhibits both clear tissue structure and good staining effect yet with the most tissue structure shrinkage. Conclusion： The three fixing agents evaluated here could all be used to fix nerve fiber while apparently acetone provides the best fixing effect.
