摘要
以活酵母细胞衍生物(LYCD)中多糖为研究对象,经sevege法脱杂蛋白、乙醇沉淀得粗多糖,由苯酚-硫酸法测定LYCD中多糖含量;该粗多糖再经SephadexG-75柱层析分离纯化得到精制;薄层层析鉴定该多糖的单糖组分;采用红外光谱对此多糖进行了初步的结构鉴定。结果表明:用sevege法脱杂蛋白3次,可得到LYCD粗多糖,其多糖含量为8.21 mg/L。该多糖中只含有一种多糖,水解得到的单糖为葡萄糖,红外图谱结果表明LYCD多糖具有多糖的一般结构。
The polysaccharide of live yeast cell derivatives (LYCD)was studied. The crude polysaccharide was precipitated with 95 % ethanol in method of ethanol precipitation and the protein was removed by Sevege method. Then the content was determined by the phenol-sulfuric acid method. The crude polysaccharide was isolated and purified by Sephadex G-75. Then the components of the product was considered by thin-layer chromatography. Finally, the structure of polysaccharide were analyzed preliminarily by infra-red spectrum. The results showed that three times were best to remove the protein. Under this condition 8.21 mg/L polysaccharides were obtained. The LYCD polysaccharides had one kind of polysaccharide which contained D-glucose after hydrolysis.The results of IR spectrum showed that LYCD polysaccharides had the general structure of polysaccharides.
