摘要
目的探讨灯盏花素脂质体肺部给药对急性肺损伤大鼠血栓调节蛋白表达的影响.方法随机将雄性SD大鼠48只分为生理盐水组、灯盏花素脂质体组、急性肺损伤组和灯盏花素脂质体干预组.检测肺组织W/D比值、MPO含量和BALF中蛋白含量,应用荧光定量PCR技术SYBR Green I荧光染料法,检测肺组织血栓调节蛋白(thrombomodulin,TM)mRNA的表达水平.结果与急性肺损伤组相比,灯盏花素脂质体干预组大鼠肺组织W/D比值、MPO含量和BALF中蛋白含量明显降低,而肺组织TMmRNA的表达水平明显升高.结论灯盏花素脂质体肺部给药能够减轻LPS致大鼠急性肺损伤的程度,促进肺组织TM的产生.
Objective To investigate the effects of expression of thrombomodulin of rats with acute lung injury. pulmonary delivery of breviscapine liposomes on Methods Healthy adult SD rats were randomly divided into control group, breviscapine liposomes group, ALI group, breviscapine liposomes +lipopolysaccharide group. Lung W/D ratio and lung MPO activity were measured, total protein concentration in BALF and the expression of TM in the lung tissue was measured by real-time RT-PCR. Results Compared with LPS group, the lung injury was evidently meliorated in breviscapine liposomes + lipopolysaccharide groupe, the lung W/D ratio was evidently lower, lung MPO activity and total protein concentration in BALF were also evidently lower, the expression of TM in the lung tissue was elevated significantly. Conclusion Pulmonary delivery of Breviscapine liposomes can meliorate the injury degree of ALI and promote the expression of TM in lung tissues.
