摘要
目的研究丙泊酚对肾移植大鼠肾脏局部血流动力学的改变和内源性线粒体凋亡信号通路中关键蛋白表达的变化。方法选择成年近交系Lewis雄性大鼠40只行异体肾移植,实验鼠分为三组:对照组(C组),肾移植组(RT组),肾移植+丙泊酚组(RT+P组)。术中分别记录实验鼠的左肾肾静脉血流速度,不同时间进行比较;用蛋白免疫印迹方法检测细胞凋亡信号通路相关蛋白Bcl-2,Bax,caspase 3和细胞色素C的表达。结果①肾静脉血流速度:RT组和RT+P组组内比较供肾在受体均比在供体显著下降(P<0.05),组间比较RT+P组供肾血流速度显著下降(P<0.05)。②凋亡相关蛋白:RT组移植肾与对照组相比Bcl-2表达减弱,Bax,caspase 3,细胞色素C表达增强;RT+P组移植肾与RT组相比Bcl-2表达增强,而Bax,细胞色素C,caspase 3表达降低。结论丙泊酚可降低肾移植大鼠移植肾的肾静脉血流速度;丙泊酚抑制大鼠移植肾脏的缺血/再灌注损伤引发的内源性线粒体细胞凋亡信号通路中相关蛋白的表达,起到肾保护的作用。
Objective To explore the effect of propofol on hemodynamics in rat renal transplantation and the key protein expression in intrinsic signaling pathways of apoptosis.Methods The models of renal ischemia/reperfusion injury during renal transplantation were established in 40 healthy adult male Lewis rats(16 donators and 16 recipients).The rats were divided into three groups:control group(C group,n=8),renal transplantation group(RT group,8 recipients and 8 donators),renal transplantation + propofol group(RT+P group,8 recipients and 8 donators).The donators'left kidneys were harvested and stored,while the recipients'left kidneys were excised.The inosculation in rats were performed between abdominal aortas,bladders and vena cava,respectively.The renal vein blood flow rates in donator kidney were recorded before excision and after reperfusion.The donator kidneys were harvested at 24 h after operation.The key apoptosis proteins,Bcl-2,Bax,cytochrome C and caspase 3 were detected with Western Blot.Results ①The renal vein blood flow rate in recipients in RT group and RT+P group significantly decreased compared with donators,respectively(P〈0.05),and the rate in RT+P group was lower than in RT group in recipients(P〈0.05).②Bcl-2 expression was lower in RT group than in C group,while the expression of Bax,caspase 3,cytochrome C was higher.Bcl-2 expression was higher in RT+P group than in RT group,while the expression of Bax,caspase 3,cytochrome C was lower.Conclusion Propofol could decrease the renal blood flow rate in renal transplantation rats.Propofol could inhibit the intrinsic signaling pathways of apoptosis induced by ischemia/reperfusion injury,and has the protective effect on kidney in rats.
