摘要
目的探讨喉鳞癌组织CDK2激酶表达与肿瘤细胞增殖之间的关系。方法取手术中获得的50例喉鳞癌组织,12例非典型增生组织和30例声带息肉组织,用免疫组化的方法检测CDK2与PCNA的表达;用流式细胞术检测喉鳞癌组织细胞周期比率。结果在喉鳞癌组织中CDK2与PCNA的阳性率表达分别为68.0%和86.0%,显著高于声带息肉组织(P<0.05);并且CDK2的表达与临床分期、病理分级、淋巴转移密切相关,与患者的年龄、性别和原发部位无关。结论喉鳞癌中CDK2过度表达可能与肿瘤细胞增殖异常密切相关。在诊断和治疗喉鳞癌中,CDK2可能是一个有重要作用的指标。
Objective To investigate the role of CDK2 in cell proliferation in laryngeal squamous cell carcinoma(LSCC). Methods The expression of CDK2 protein and proliferating cell nuclear antigen(PCNA)was analyzed in 50 specimens of LSCC,12 specimens of atypical hyperplasia and 30 specimens of vocal cord polyp obtained during operation by immunohistochemical method.The ratio of cell cycle was determined in LSCC using flow cytometry. Results The positive rates of CDK2 and PCNA in LSCC were 68.0%(34/50) and 86.0%(43/50),significantly higher than those in the vocal cord polyp(P〈0.05).Expression of CDK2 in LSCC was positively related with expression of PCNA and cell proliferation.CDK2 expression was closely related with tumor grade,pathological grade and lymphatic metastasis(P〈0.05),but not with primary site of LSCC,gender and age. Conclusion The results indicate that overexpression of CDK2 may be strongly correlated with abnormal proliferation in LSCC,and it may be an important marker in the diagnosis and treatment of LSCC.
