摘要 为进一步了解抗肿瘤药物5-氟尿嘧啶(5-fluorouracil,5-Fu)的作用机制,研究了5-Fu对体外培养的人乳腺癌细胞系(MCF-7)和人宫颈癌细胞系(Hela)这两种肿瘤细胞生长繁殖的影响及有效的诱导肿瘤细胞凋亡的浓度和作用时间。使用Leukostat染色法测定了细胞凋亡的百分率;DNA电泳分析了细胞凋亡出现的特异的DNAladder现象。结果表明,用10~20mmol/L浓度范围的5-Fu作用24h,能够引起肿瘤细胞产生典型的凋亡现象。 This study detected the effective concentrations of 5 -fliorouracil and treatment time to induce the apoptosis of the MCF -7 cells and the Hela cells in vitro culture in order to understand the mechanism of anti - tumor medicine 5 - fluorouracil, the cell apoptosis rate was counted by Leukostat staining. The DNA fragments of the apoptosis cells was analyzed by means of agarose electrophoresis. The results suggested that after MCF -7 and Hela cells were treated with 10 - 20mmol/L of 5 - Fu for 24 h, the characteristic phenomenon of apoptosis was induced.
