摘要
目的:建立一种简便、快速的高效液相色谱(HPLC)荧光检测法测定人体内血浆中双嘧达莫的浓度。方法:采用Kro-masil C18柱(4.6 mm×150 mm,5μm),柱温30℃,流动相为0.02 mol.L-1磷酸二氢钾(含0.04%的三乙胺)-乙腈(39:61),流速为1.0 ml.min-1,荧光检测激发波长290 nm,发射波长485 nm。氟伐他汀为内标。结果:双嘧达莫在10~4000μg.L-1范围内线性关系良好(r=0.9997)。日内和日间RSD%均低于10%。结论:该方法灵敏度高、快速、简便、无杂质干扰,适合于双嘧达莫的血药浓度监测及药物动力学研究。
Objective: To establish a simple and rapid method for determination of dipyridamole in human plasma by HPLC with fluorescence detection. Methods: Chromatographic column was Kromasil C1s (4.6 mm '150 mm,5 μm) and 30℃ of column temperature. The mobile phase consisted of 0.02 mol·L^-1 potassium dihydrogen phosphate(containing Triethylamine 0.04%)- acetonitrile (39:61) with a flow rate of 1.0 ml·min^-1.A fluorescence detector was set atλcx290nm,λcm485 nm. Fluvastatin was used as internal standard. Results: The calibration curve was in good linearity over the range of 10-4000 μg·L^-1 (r=0.9997).The intraday and inter-day RSD were less than 10%. Conclusion: The method is sensitive, rapid and suitable for the determination of dipyridamole in plasma.
