摘要 目的观察全反式维甲酸(all-trans-retinoic acid,ATRA)对甲状腺鳞癌细胞株SW579诱导分化作用中细胞增殖的影响。方法以终浓度为10^(-7)、5×10^(-7)、10^(-6)、5×10^(-5)、10^(-5)mol/L的全反式维甲酸作用SW579细胞株,在倒置显微镜下观察细胞生长情况及形态变化;采用MTT比色法测定细胞存活率;流式细胞仪检测细胞周期。结果全反式维甲酸能使细胞趋向良性分化,抑制了肿瘤细胞增殖,使细胞滞留在S期。结论全反式维甲酸对甲状腺鳞癌细胞株SW579有剂量依赖性的增殖抑制作用,并且在低浓度时(10^(-7)、5×10^(-7)、10^(-6) moL/L)可能对细胞具有诱导分化作用。 Objective To observe the influences of all - trans - retinoic acid (ATRA) on proliferation of of thyroid squamous cell carcinoma cell line (SW579) in inducing differentiation in vitro. Methods Treated with different doses of ATRA ( final concen- tration of 10^-7, 5 ×10^-7, 10-6, 5 ×10^-6, 10^-5mol/L) , the cells were observed With an inverted microscopca convert microscope in shape; The the proliferation of the cells was observed with MTT and the cell cycle was measured by flow cytometry . Results Af- ter cells were treated with ATRA, the malignant phenotypes of cells gradually changed to the mature cells. The proliferation index de- creased markedly. The cell cycle was arrested at S stage. Conclusions ATRA can inhibit the proliferation of the cell line SW579 with a dose -dependent manner, and maybe the thyroid cancer cells were induced differentiation in low concentration (10^-7mol/L, 5 ×10^-7mol/L, 10^-6mol/L).
