睾丸固定术对隐睾大鼠生殖细胞凋亡与eNOS表达的影响 被引量:1

Assessment of Germ Cell Apoptosis and Enos Expression After Scrotal Replacement of Experimentally Cryptorchid Rat Testis

摘要 目的探讨睾丸固定术对隐睾大鼠生殖细胞发育与凋亡的影响。方法22日龄SD雄性大鼠复制左侧隐睾模型。实验随机分为假手术组、隐睾组和隐睾固定术组,每组10只。建立左侧隐睾模型,隐睾固定术组于建立左侧隐睾模型后14d行隐睾下降固定术。以生物素-dUTP/酶标亲和素测定法检测睾丸生殖细胞凋亡数目,用免疫组化SABC法检测eNOS基因表达。结果与假手术组相比,隐睾组和隐睾固定术组隐睾侧睾丸发生生精细胞凋亡的数目显著增加(P<0.01)。与隐睾组相比,睾丸固定术后生殖细胞凋亡明显减少(P<0.01)。隐睾组睾丸退化的生精细胞胞浆中eNOS基因表达明显增强。结论隐睾大鼠生殖细胞凋亡增多,睾丸固定术可降低生精细胞的凋亡水平。eNOS表达与生精细胞凋亡有密切关系。 Objective This thesis aims to explore the effect of orchidopexy on germ cell development and apoptosis in rat model. Methods Thirty male SD rats (22 - day old) were randomly divided into three groups: left - sham - operated group ( n = 10,) , cryptorchid group (n = 10), orchidopexy group (n = 10) . Rats were rendered unilaterallycryptorchid, and 14 days later, orehidopexy was performed on a subset of these rats. Forty days after the initial procedure, testes were harvested for immunohistochemical stud- ies. Apoptosis was detected by in situ 39 - end - labeling of DNA with digoxigenin - dUTP, and eNOS protein was detected using an eNOS monoclonal antibody. Results The number of apoptotic germ cell in cryptorchid testes increased observably compared with the left - sham - operated group ( P 〈 0.01 ) , and the number of apoptotic germ cell in orchidopexid testes decreased observably compared with cryptorchid testes (P 〈 0.01 ). Conclusions Cryptorchidism increases germ cell apoptosis, and orchidopexy can lower the level of germ cell apoptosis, eNOS expression is closely related to germ cell degeneration.
