摘要
目的研究钙蛋白酶(calpain)在正常豚鼠耳蜗中的定位与表达。方法应用免疫组织化学SABC法结合显微图像分析技术,观察耳蜗组织中calpain的表达;同时采用电生理指标听性脑干反应(auditory brainstem response,ABR)检测豚鼠的听力。结果calpain免疫反应活性主要见于耳蜗血管纹、螺旋韧带、螺旋神经节及毛细胞,螺旋神经节的染色较深;其中,calpain 1表达多见于螺旋神经节细胞的胞浆中,而calpain 2免疫染色几乎出现在整个螺旋神经节细胞中。结论calpain在正常豚鼠耳蜗的多种细胞中都有表达。
Objective To investigate the localization and expression of calpains in the cochlea of normal guinea pig. Methods The expression of calpains was detected by the SABC method of immunohistochemistry combined with imaging analysis technique. Auditory brainstem response (ABR) measurement was used to test the hearing of guinea pig. Results Immunoreactivity for calpains was mainly found in stria vascularis, spiral ligament, spiral ganglions and hair cells, and immunostaining was greater in spiral ganglion. Furthermore, the calpain 1 expression dominantly existed in the cytoplasm of spiral ganglions, while calpain 2 immunostaining was almost found in the whole spiral ganglion cells. Conclusions Calpain exists in many cell types of normal guinea pig's cochlea.
