摘要
目的观察乙酸镁对心肌缺血-再灌注损伤过程中心肌保护作用及其机理。方法取50只SD大鼠随机分为5组:假手术组(Sham),模型对照组(Control),乙酸镁低剂量组(50mg/kg)、乙酸镁中剂量组(70mg/kg)、乙酸镁高剂量组(100mg/kg)。采用整体大鼠心肌缺血-再灌注损伤模型,结扎冠状动脉左前降支30min后,再灌注60min,测定心肌组织SOD活性、MDA含量及血清中LDH和镁离子含量;观察ADP诱导的血小板聚集功能的变化。结果与模型组相比,乙酸镁三个不同剂量组血中Mg2+浓度升高,LDH和MDA含量明显降低,SOD活性显著升高;抑制ADP诱导的血小板聚集,以上作用都呈剂量依赖性。结论乙酸镁对大鼠心肌缺血再灌注损伤有保护作用,其机制与抗ADP诱导的血小板聚集,减轻氧自由基损伤,改善心肌细胞能量代谢有关。
Objective To observe the cardioproteetive effect and mechanisms of magnesium acetate on myocardial ischemia reperfusion injury in rats. Methods 50 SD rats were randomly divided into five groups: Sham, Model, low-dosage magnesium aeetate(50mg/kg), media-dosage magnesium acetate (70mg/kg), high-dosage magnesium acetate (100mg/kg). The model of myocardial ischemia reperfusion in rats were made by deligating then releasing the suture advanced around the left coronary artery inducing 30 minutes ischemia and 60 minutes reperfusion. When the reperfusion is executed, blood samples were collected for assaying contents of LDH and contents of Mg2 + ;meanwhile heart samples were obtained for determining the activities of SOD and contents of MDA; anticoagulated blood samples were collected for assaying the change of platelet aggregation (AGG) induced by ADP. Result: Compared with Model, the three agents all reduced the contents of MDA and LDH; simultaneously increased the activities of SOD and the content of Mg2+; inhibited the platelet aggregation induced by ADP. The dosage dependence was observed of the effect mentioned above. Conclusion Magnesium acetate has protective effects on myocardial ischemia reperfusion injury. The protective effect may be related to inhibiting platelet aggregation induced by ADP, clearing oxygen free radical and improving myocardial cell energy metabolism.
