摘要
目的探讨解偶联蛋白2(UCP2)在大鼠肝脏组织中的表达及其与年龄的关系。方法选用健康雄性SD大鼠,按年龄分为新生组、成年组和老年组(n=10),实验室适应性喂养7d后断头处死,分别取各组大鼠肝组织测定丙二醛、谷胱甘肽、ATP的含量并采用Westernblot方法检测肝细胞线粒体内UCP2的表达并进行定量分析。结果实验证实老年组大鼠肝组织中MDA(丙二醛)含量最高,GSH(谷胱甘肽)和ATP含量最低;老年组UCP2的水平明显高于新生组和成年组。老年组与新生组比较P〈0.01;新生组与成年组比较P〈0.01;老年组与成年组比较P〈0.01。各组别比较均有显著性差异。结论UCP2在肝脏组织对肝细胞的损伤与衰老过程中起重要作用。
Objective To discuss the expression of UCP2 in the livers of rats. Methods Thirty healthy male SD rats were chosen and divided into three groups: newly - born group, adult group and old - age group, which were fed for 7 days and then killed by cutting heads. These rats hepatic tissue was taken out respectively to test the content of MDA, GSH and ATP, and then test the expression of UCP2 in the mitochondrion of liver cells in the way of Western blot. Results According to the experiment, the content of MDA in the hepatic tissue of old - age group is the highest, and the content of GSH and ATP is the lowest; The quantities of UCP2 of old - age group is distinctively more than newly -born group and adult group. In the comprison of old- age group and newly- born group, P 〈 0.01; In the comprison of newly- born group and dult group, P 〈 0.01; In the comprison of old- age group and adult group, P 〈 0.01. The comparisons of every two groups have distinctive difference. Conclusion UCP2 plays an important role in the procedure of injury and senescence of liver cell in hepatic tissue.
