针刺对吗啡戒断大鼠情绪及氨基酸水平的影响 被引量:7

摘要 目的探讨针刺对吗啡戒断大鼠情绪的作用及对蓝斑(locus ceruleus,LC)氨基酸水平的影响。方法采用剂量递增方法,皮下注射吗啡形成大鼠依赖后,给予纳洛酮催促,建立吗啡戒断后焦虑模型。采用"神门穴"针刺治疗和十字迷宫行为学测试的方法,观察针刺对吗啡戒断后大鼠的焦虑情绪的作用;采用高效液相色谱-电化学检测法测定甘氨酸(glycine,Gly)、谷氨酸(glutamate,Glu)和γ-氨基丁酸(γ-aminobutyric acid,GABA)含量。结果①针刺可使吗啡戒断后焦虑模型大鼠在开臂滞留时间百分比增大(P<0.01),探究行为增多(P<0.01)。②戒断大鼠蓝斑Gly、GABA含量降低,Glu含量升高(P<0.05);针刺使戒断大鼠蓝斑Gly、Glu含量增高,Glu含量降低(P<0.05)。结论针刺对吗啡戒断后大鼠的抗焦虑效应可能与针刺调节焦虑模型大鼠脑内Gly、GABA、Glu的释放有关。 Objective To observe the impact of acupuncture on the anxiety and level of amino acids within locus ceruleus (LC) in morphine withdrawal rats. Methods Morphine addictive model was constructed by dose-increasing administration. And the rats were given naloxone to establish anxiety model of morphine withdrawal. To observe the effect of acupuncture on "shenmen point" on with- drawal rats'anxiety. The elevated plus-maze test was used to assess the rat's anxiety. High performance liquid chromatography-masss pectrometry( HPLC-MS} was used to quantify the Gly and GABA,Glu in LC. Results (1)The OT% in open arms went up and exploring behavior increased on withdrawal rats' anxiety after acupuncture ( P 〈 0.01 ). (2)Quantity of Gly and GABA within LC decreased and Glu increased in morphine withdrawal rats( P 〈0. 05 ). level of Gly,GABA increased and Glu decreased after acupuncture on withdrawal rats ( P 〈 0. 05 ). Conclusion The effect of acupuncture anti-anxiety applied on the morphine withdrawal rats was possibly related to the release of Gly,GABA,Glu by acupuncture in brain of anxiety rat's.
