摘要
目的探讨手掌参醇提物(Gymnadenia conopsea Alcohol Extract,GcAE)对染矽尘大鼠肺组织肿瘤坏死因子-α(Tumor Nec-rosis Factor-α,TNF-α)表达的影响。方法将Wistar大鼠随机分为对照组、染矽尘组、GcAE治疗组,每组40只。采用气管暴露法建立大鼠矽肺模型。灌胃给予GcAE(8g.kg-1.d-1)治疗后,在第7、14、21、28和60天时每组分别随机取8只大鼠宰杀,制作大鼠肺组织芯片,用免疫组织化学PV法检测组织芯片TNF-α的表达,用Image-Pro Plus Version 4.5 for windowsTM图像分析系统对TNF-α的表达做定量分析。结果矽尘组TNF-α的积分光密度各时间点均高于对照组,在第7、14、21、28天时有统计学意义(P<0.01)。手掌参组各时间点肺组织TNF-α的积分光密度低于染矽尘组,在第7、14、21、28天时有统计学意义(P<0.01)。结论GcAE能够明显抑制矽尘诱导的大鼠肺组织TNF-α的过度表达。
im To explore the effects of C, cAE on TNF-α expression of lung in rats induced by silica. Methods One hundred and twenty Wistar rats were randomly divided into control group, silica group and GeAE-treated group. Silicotic animal models were established by intratracheal infusion of silica dust suspension. From the second day of model establishment, rats in GcAE-treated group were o- rally given GcAE (8 g/kg, corresponding to raw herb) everyday. At 7,14,21,28,60 days after establishment of the animal model, eight rats in each group were sacrificed and samples were collected. The expression of TNF-α protein in paraffin-embedded lung sections were stained immunohistochemistrily on tissue microarray with PV method. Results TNF-α positive area percent of the lung was increased in silica group compared with the corresponding control group, and significantly at 7,14,21 and 28 days (P 〈 0. 01 ). Treated with GcAE can reduce such enhancement at all time points, and significantly at 7,14,21 and 28 days ( P 〈 0. 01 ). Conclusion GcAE plays the role of intervention through inhibiting the over expression of TNF-α in lung tissue of rats induced by silica.
