人脑脊液中利多卡因的高效液相色谱法测定 被引量:5

摘要 目的建立检测人脑脊液中利多卡因的方法。方法采用RP-HPLC法测定人脑脊液中药物浓度,色谱柱为Lichrospher C18(4.6×250 mm,5μm),流动相为甲醇-水-冰醋酸-三乙胺(62∶38∶0.3∶0.6),流速1 mL.min-1,检测波长220 nm。结果在0.1-8 mg·L^-1范围内利多卡因峰面积与内标峰面积之比与浓度线性关系良好,最低检测浓度为0.03 mg·L^-1,回收率利多卡因为93.77-95.64%,日内、日间RSD均小于10%。结论本测定方法灵敏、准确、简便,适合于对利多卡因的临床使用监测及药代动力学研究。 Objective To develop a simple,sensitive and rapid method for the determination of lidocaine in human cerebrospinal fluid.Methods The concentration of lidocaine was determined by RP-HPLC method,the analysis involved a 250 mm × 4.6mm column packed with C18(5 μm),the mobile phase consisted of methyl alcohol-water-glacial acetic acid-triethylamine(62∶38∶0.3 ∶0.6),dertermination wave was 220 nm.Results A good linearity was obtained in the range of 0.1~8 mg·L^-1,the recovery of lidocaine was between 93.77-95.64%,the relative stand deviations of within-day and between day were less than 10%,the minimal detectable concentration in plasma was 0.03 mg·L^-1.Conclusion The result demonstrated that the method was sensitive,accurate and simple for the determination of lidocaine in human cerebrospinal fluid,it was suitable for the pharmacokinetics study and clinical monitoring of lidocaine.
