滋阴活血解毒中药对血管内皮细胞损伤的保护作用 被引量:11

摘要 目的观察滋阴活血解毒中药对葡萄糖(Glu)、胰岛素(Ins)、氧化低密度脂蛋白(ox-LDL)诱导的人脐静脉内皮细胞损伤的干预作用及其保护机制。方法以人脐静脉内皮细胞株ECV-304为受试对象,以一定浓度的Glu、Ins、ox-LDL诱导体外血管内皮细胞损伤,同时观察滋阴活血解毒中药对该细胞损伤的保护作用。各实验组细胞继续培养48 h后,分别检测细胞活性及细胞培养液中组织型纤溶酶原激活物(t-PA)、纤溶酶原激活物抑制物(PAI)、细胞粘附分子(ICAM-1)的含量。结果细胞培养48 h后,模型组较正常组细胞活力明显下降(P<0.01),PAI及ICAM-1含量明显升高(P<0.01),而不同剂量的滋阴活血解毒中药能显著改善细胞活力(P<0.05),降低PAI及ICAM-1含量(P<0.01),对t-PA的影响无显著性意义。结论滋阴活血解毒中药对Glu、Ins、ox-LDL诱导损伤的血管内皮细胞有保护作用,而这一作用可能是通过促进内皮细胞的纤溶功能、抑制与炎症反应相关的细胞粘附过程等途径实现的。 Objective To investigate the effects of Ziyin Huoxue diedu Chinese herbs (ZYHXJD) on vascular endothelial cells injured by glucose, insulin and low-density lipoprotein. Methods Human umbilical vein endothelial cell line ECV-304 was exposed to different concentration of glucose, insulin and oxidized low-density lipoprotein (Ox-LDL), and the effects of ZYHXJD on the injured vascular endothelial cells were investigated. The cells in each group were cultured for additional 48 h. And then, cell viability was examined, and the supernatants were used to determine the contents of tissue plaminogen activator (tPA), plasminogen activator inhibitor (PAI), and intercellular adhesion molecule-1 (ICAM-1) respectively. Results The viability of the cells in the model group decreased markedly (P〈0.01), and the secretion of PAI and ICAM-1 in the model group was significantly higher than that of the blank group (P〈0.01). After treating with different dose ZYHXJD, the cell viability was significantly higher than that of the model group (P〈0.05). The cells PAI and ICAM-1 were significantly lower than those of the model group (P〈0.01). Conclusion ZYHXJD have the protective function to the vascular endothelial cells, and this function might be realized by advancing fibrinolysis and inhibiting intercellular adhesion relating with inflammatory reaction.
