摘要
目的探讨HGF联合EGF诱导大鼠骨髓间充质干细胞(MSCs)后对急性肝脏损伤修复。方法D-氨基半乳糖腹腔注射复制大鼠肝脏损伤模型,分为实验组和对照组,实验组用HGF联合EGF诱导后的MSCs对损伤的大鼠门静脉移植,对照组用未诱导MSCs对模型行门静脉移植,分别于1、3、7、14d处死查生化(丙氨酸氨基转移酶ALT、天冬氨酸氨基转移酶AST、总胆红素TBi)、切片行常规病理。结果MSCs门静脉移植结果,肝功能比较:肝损伤模型的肝功指标均明显异常,两组均出现检测指标逐步下降低的趋势,但在同时间段内,实验组下降幅度更大。对照组与实验组比较:大鼠肝脏各项生化指标除第1天无显著变化(P>0.05)外,其余各时间点差异均有统计学意义(P<0.05)。病理切片显示:实验组肝细胞变性坏死程度明显轻于对照组,7、14d尤为明显。结论将诱导后MSCs移植给受损伤的肝脏,能在一定程度上修复肝脏的损伤,能挽救损伤的肝功能。
Objective To investigate the possibility of the rat mesenchymal stem cells (MSCs) induced by hepatocyte growth factor plus E growth factor to provide a seed cell for cell transplantt to cure all kinds of acute hepatic injury. Methods D - galactosamine was injected to abdominal cavity in the rat so as to duplicate liver injury model. Rat was divided into two groups, the experimental group and control group. In the experimental group,mesenchymal stem cells induced by HGF plus EGF, were transplanted into injured portal vein in the rat; in the two group, mesenchymal stem cells were transplanted into injured portal vein in the rat. After rats were put to death on 1,3, 7 and 14 d respectively, ALT,AST , TBiL and ALP were detected in the blood; section was proceeded to routine pathology. Result Blood biochemistry results show that there is different significance in the two groups. Pathological findings display that in the experimental group hepatic degeneration and necrosis level is obviously lighter than that in the control group , in particular, at 3 and 4 days. Condusion In addition to , when induced mesenchymal stem cells are transplanted into injured liver, to some extent, they can save the liver and repair the injured liver function.
