复方瓜子金颗粒中秦皮乙素、蒙花苷和polygalasaponin ⅩⅪ的HPLC-DAD-ELSD法测定 被引量:4

摘要 目的:建立一种同时测定复方瓜子金颗粒中秦皮乙素、蒙花苷和 polygalasaponin XXI 含量的 HPLC-DAD-ELSD 方法。方法:采用 HPLC-DAD—ELSD 法,以 Diamonsil C_(18)(250 mm×4.6 mm,5 μm)为色谱柱,乙腈(A)-0.05%三氟醋酸水溶液(B)为流动相,梯度洗脱[0~10 min(15%A),10~30 min(15%A→28%A),30~40 min(28%A),40~50 min(28%A→32%A)],流速为1.0 mL·min^(-1),二极管阵列检测器的检测波长为340 nm,蒸发光散射检测器的漂移管温度为105℃、载气流速为2.6 L·min^(-1)。结果:秦皮乙素、蒙花苷和 polygalasaponin XXI 的线性范围分别为4.75~118.75μg·mL^(-1)(r=0.9998),4.02~100.50μg·mL^(-1)(r=0.9999),19.62~157.00μg·mL^(-1)(r=0.9993);加样回收率(n=9)分别为102.7%,100.9%,97.3%。结论:测定方法准确、重现,可为复方瓜子金颗粒的质量控制提供借鉴。 Objective:To establish a method for the determination of aesculein,linarin and polygalasaponin XXI in compound Guazijin granules simultaneously. Methods:An HPLC - DAD - ELSD assay was performed on Diamonsil C18 column(250 mm×4.6 mm, 5μm) with acetonitrile (A) - 0.05 % trifluoroacetic acid in water (B) as mobile phase by a gradient elution program [ 0 - 10 min ( 15 % A ), 10 - 30 min ( 15 % A→28 % A), 30 - 40 min ( 28 % A ), 40 -50 min(28%A→32%A) ] at a flow rate of 1.0 mL · min^-1. The detection wavelength of DAD was 340 nm and the evaporator tube temperature of ELSD was set at 105℃ with the nebulizing gas flow - rate of 2.6 L · min^-1. Results:The linear ranges of aesculein,linarin and polygalasaponin XXI were 4.75 - 118.75 μg · mL^-1( r =0. 9998) ,4.02 - 100.50 μg · mL^-1(r = 0. 9999) and 19.62 - 157.00 μg · mL^-1(r = 0. 9993), respectively. The recoveries were 102.7% , 100.9% and 97.3% ,respectively. Conclusions:The method is found to be accurate and reproducible for simultaneous analysis of aesculein,linarin and polygalasaponin XXI in compound Guazijin granules, and provides a reliable way for evaluating the quality of it.
