川芎嗪对高糖环境大鼠肾小球系膜细胞增殖及其细胞外基质含量的影响 被引量:6

摘要 目的:探讨川芎嗪对高糖环境下大鼠肾小球系膜细胞(MCs)增殖及其细胞外基质含量的影响。方法:体外培养大鼠肾小球系膜细胞株,分对照组、高糖组、TMP组,分别培养12h、24h、48h,CASY细胞计数仪计数细胞存活率;以CCK-8法检测细胞增殖;以ELISA法检测细胞上清液细胞外基质ColIV及FN的含量。结果:各TMP浓度组之间存活率无统计学差异(P>0.05);与对照组比较,高糖下培养24h、48hMCs增殖显著增快(P<0.01),分泌的ColIV及FN含量增多(P<0.05);与高糖组比较,TMP各组MCs增殖显著减慢(P<0.01),分泌ColIV及FN减少(P<0.01或P<0.05)。结论:川芎嗪可以有效抑制高糖的促增殖作用并能减少系膜细胞外基质的增加。 Objective: To explore the effects of tetramethylpyrazine on proliferation of mesangial cells and the concentration of extracellular matrix in high glucose cultured rat mesangial cells. Methods: Rat mesangial cells were divided into control group, high glucose group and TMP group, and were cultured for 12 h, 24 h and 48 h. At indicated time points, cellular survival rate and proliferation were identified with cell counting instrument and CCK-8, The supernatants were collected and the Collagen type IV and Fibronectin were detected with enzyme-linked immunosorbent assay. Results: The survival rates of different groups were not involved (P〉0.05). Compared with control group, the cellular proliferation and the concentration of Collagen type IV and Fibronectin increased in high glucose group at 24 h and 48 h (P〈0.05). Compared with that in high glucose group, the cellular proliferation and the concentration of Collagen type IV and Fibronectin declined in different concentration TMP groups (P〈0.01,P〈0.05). Conclusion: Tetramethylpyrazine can significantly inhibit proliferation and secretion of Collagen type IV and Fibronectin in high glucose induced rat mesangial cells.
