摘要
目的:探讨大鼠颅脑损伤后Survivin蛋白表达和Caspase-3活性变化的规律和意义。方法:42只Wistar大鼠随机分为对照组(n=6)与损伤组(n=36)。采用自由落体法建立大鼠颅脑损伤模型,分别于伤后不同时间点,应用免疫组织化学法检测Survivin和Caspase-3蛋白表达变化,利用分光光度法测定Caspase-3活性变化,应用流式细胞仪检测神经细胞凋亡情况。结果:与对照组相比,Caspase-3活性、细胞凋亡率及Caspase-3、Survivin蛋白表达在大鼠脑损伤后均显著升高(P<0.01)。通过相关性分析,大鼠脑损伤后脑组织Caspase-3的活性与细胞凋亡率具有正相关性(P<0.01),而和Survivin表达具有负相关性(P<0.01)。结论:大鼠颅脑损伤后,Survivin表达增加可下调Caspase-3的活性,减少细胞凋亡。
Objective: To investigate the dynamic change and significance of Survivin protein expression and Caspase -3 activation following traumatic brain injury (TBI) in rats. Methods: Forty -two adult Wistar rats were randomly divided into trauma group ( n = 36 ) and control group ( n = 6 ). The experimental TBI model was established by Feeney's method. Tissue samples were harvested at different time points after injury for inspecting the expression of Survivin and Caspase -3 protein by immunohistochemical method, determining the Caspase -3 activity by colorimetric assay and measuring apoptosis by flow cytometer. Results:Compared to the control group, Caspase - 3 activation, the number of apoptotic cell, and the Survivin and Caspase - 3 expressions in trauma group were all increased significantly (P 〈0. 01 ). According to the correlation analysis, the activity of Caspase-3 was positively correlated with apoptotic rates (P 〈0. 01 ), but negatively correlated with the Survivin expression (P 〈 0.01 ). Conclusion: The increasing expression of Survivin after TBI may down-regulate Caspase- 3 activation and inhibit apoptosis.
