摘要
目的探讨Caspase-3 mRNA在小鼠肾小体发育中表达的变化特征及其可能的生物学意义。方法选取胚龄14d、16d、18d胎鼠和出生后1d、3d、7d的仔鼠肾组织,采用原位杂交技术及体视学分析方法对Caspase-3 mRNA在不同发育阶段肾小体中的表达进行定性及定量观察。结果Caspase-3 mRNA在小鼠肾小体发育的不同阶段都有阳性表达。在Ⅰ期和Ⅱ期肾小体的表达较低,进入Ⅲ期肾小体时,其表达达高峰,Ⅳ期肾小体中的表达稍有下降,Ⅴ期肾小体中的表达显著降低,其体积密度分别为12.35±3.78(%)、15.63±4.05(%)、43.28±6.09(%)、33.45±5.32(%)和10.28±2.56(%)。结论Caspase-3 mRNA在小鼠不同发育阶段肾小体内的表达不同,它对肾小体的正常发育可能有重要意义。
Objective To investigate the expression of caspase-3mRNA during the development of renal corpuscle in mouse and its potentially biological significance. Methods The kidney of embryonic 14, 16 and 18 days and postnatal 1, 3, 7 days mice was taken in this study. In situ hybridization and stereological methods were used to observe and measure the expression of caspase-3 mRNA in developing mouse renal corpuscle. Results The caspase-3 mRNA positive cells can be seen in all stages of the developing renal corpuscle. The expression of caspase-3 mRNA was lower in Ⅰ and Ⅱ stages, the highest in Ⅲ stage, then gradullaly decreased in Ⅳ stage, and less in Ⅴ stage. The volume density of five stages of developing renal corpuscle was 12.35 ± 3.78(%), 15.63 ± 4.05(%), 43.28 ± 6.09(%), 33.45 ± 5.32(%) and 10.28 ± 2.56(%) respectively. Conclusion The caspase-3 mRNA expressed differently in every stage of developing renal corpuscle and probably play a significant role during the development of the renal corpuscle in mouse.
