摘要
目的探讨IL-1β和TNF-αmRNA在大鼠实验性骨关节炎软骨表达的变化和意义。方法采用Hulth法制作大鼠膝关节OA动物模型,于术后8周取标本并观察关节软骨形态,RT-PCR技术检测mRNA表达。结果OA模型侧关节软骨的IL-1β和TNF-αmRNA表达均高于对照侧。结论OA软骨中存在IL-1β和TNF-αmRNA表达的上调,可能是OA的发病机制之一。
Objective To explore the expression and significance of cytokine IL-1β and TNF-α mRNA in rat cartilage with experimental osteoarthritis. Methods The animal models of experimental OA in rat knee joint were established with Hulth method. The articular cartilages were harvested after 8 weeks. Morphological changes were found through observing the appearance of cartilage and the expression of mRNA was detected through RT-PCR techniques. Results The expression of IL-1β and TNF-α mRNA in OA models was statistically more than that in controls. Conclusion The up-expression of IL-1β and TNF-α mRNA might be a possible mechanism of OA.
