摘要
目的:检测大鼠坐骨神经损伤后降钙素基因相关肽(CGRP)在运动终板中的变化,同时观察运动终板的变化,以探讨CGRP与运动终板退变的关系。方法:切断大鼠坐骨神经并结扎其近端,建立周围神经损伤模型,分别于损伤后1、2、3、4、6、8、10和12周取胫前肌肌腹标本,分别以免疫组化法检测CGRP,镀银染色法进行运动终板染色。结果:损伤1周后运动终板中CGRP立即完全消失,镀银法显示运动终板1、2周无明显变化,3周开始染色浅淡、不均匀,8周后运动终板完全消失。结论:(1)CGRP与运动终板的退变相关;(2)CGRP检测法能更早显示运动终板的改变,灵敏度高。
Objective:To inquire the relationship of calcitonin gene related peptide(CGRP) and degeneration of motor end plate(MEP) Method: Sciatic nerves were cut and their proximal segments were ligated to prepare the model of peripheral nerve injury The venter of the muscle tibialis anterior were chosen at 1、2、3、4、6、8、10、12 weeks postoperatively to carry out the experiment The change of CGRP in the MEP was investigated after sciatic nerves injury with immunohistochemical method in present study Moreover,the degeneration of MEP was observed with traditional silver staining method Result:CGRP was completely disappeared promptly in the MEP after 1 and 2 weeks. There was not obvious change in the MEP after 1 and 2 weeks; at the 3 weeks postoperatively,the staining was light and sparse 8 weeks later,the MEP completely disappeared Conclusion:(1)CGRP was related to degeneration of MEP;(2)CGEP investigated method could show the change of MEP earlier and had high sensitivity
