摘要
目的:检测P450芳香化酶(P450A)蛋白和mRNA在子宫内膜异位症中的表达,并探讨其在发病中的作用。方法:采用逆转录聚合酶联反应(RT-PCR)和免疫组化技术检测45例卵巢巧克力囊肿的囊壁(巧囊组)、32例卵巢内异症的在位内膜(在位内膜组)和35例正常子宫内膜(正常对照组)中P450AmRNA及蛋白的表达。结果:巧囊组P450A mRNA表达量高于在位内膜组(P<0.01);35例正常子宫内膜中仅有2例呈阳性表达;免疫组化检测巧囊壁P450A蛋白其表达特征与mRNA一致。结论:子宫内膜异位症异位灶中有P450A表达,这种雌激素的合成酶能在异位灶局部合成异常的雌激素;卵巢内异症的在位内膜有P450A表达,提示在位内膜本身局部有雌激素的合成,其保护机制劣于正常内膜。子宫内膜异位症中P450A的表达在其发病中起到重要作用。
Objective To detect the expression of P450 aromatizing enzyme (P450A) protein and messenger RNA (mRNA), and discuss the effection in endometriosis. Methods PT-PCR and immunohistochemistry method was used to detect the expressions of P450A and mRNA in 112 subjects, including 45 cases of ovarian endometriosis, 32 cases of ovarian eutopic endometrium and 35 cases of normal endometrium. Results P450A mRNA levels were higher in the ectopic endometrium than that in eutopic endometrium (P 〈 0.01 ) ; P450A mRNA was present in only 2 of 35 normal uterine endometrium samples. The expression characteristic of P450A protein was the same as one of P450A mRNA in ectopic endometrium. Conclusions The abnormal expression of P450A may lead to local high estrogen biosynthesis in endometriosis. The expression of P450A in eutopic endometrium demonstrated that the protective mechanism of eutopic endometrium was worse than that of normal endometrium. The abnormal expression of P450A probably plays an important role in the development and growth of endometriosis.
