贵州野生淡黄花百合离体快繁研究

Study on in vitro Rapid Propagation of Lilium sulphureum

[目的]为获得贵州野生淡黄花百合的离体快繁体系。[方法]以MS为基本培养基,研究了影响淡黄花百合珠芽不定芽诱导、小鳞茎增殖和生根培养的主要因素。[结果]在淡黄花百合初代培养过程中,生长素浓度、珠芽大小、珠芽处理方式、接种时间均能对不定芽的诱导产生影响。其中,以8月下旬采取粒径大于等于1 cm的珠芽,剥去外层鳞片后接种在附加0.1 mg/LIBA的1/2 MS培养基上的不定芽的诱导效果最好。培养基类型和继代时间对增殖培养有显著影响,以40 d内转接继代在1/2 MS+0.1 mg/LKT+0.1 mg/LIBA的培养基上的增殖效果最好,其愈伤组织诱导率达97.3%,小鳞茎诱导率高达100%,且不定芽生长健壮。在1/2 MS+0.1 mg/L IBA培养基上生长和生根情况均为良好。按照培养室炼苗、室外炼苗和移栽的程序,淡黄花百合试管苗移栽成活率在98%以上。[结论]成功实现了淡黄花百合的驯化移栽,该技术体系可以为贵州野生淡黄花百合的资源保护和资源利用提供参考。

[Objective] The research aimed to acquire the rapid propagation of Lilium sulphureum.[Method] taking MS media as basic media,the major factors which affected inducement of adventitious buds,multiplication of bulbs and radication of adventitious buds were studied.[Result] The hormone concentration,the diameter and the treating mode of bulbils and the inoculation time all effected on the induction of adventitious buds in first culture of Lilium sulphureum.It was the best to the induction of adventitious buds that the explants whose diameter was bigger than 1 cm,were taken in the last ten-day of August,after shucking off the outer squamas,and the bulbils were inoculated on the 1/2 MS＋0.1mg/L IBA.The media types and second culture period all affected the multiplication of Lilium sulphureum.It had the best effect on multiplication to take 1/2 MS＋0.1mg/L KT ＋0.1mg/L IBA as multiplication media and second culture every 40 d.In this way,the callus induction rate was 97.3%,the percentage of bulb induction was 100%,and the adventitious buds grew vigorous.In the 1/2 MS＋0.1 mg/L IBA media,the radication was well.According the process of domestication in culture room and outdoors,then transplanting the seedlings,the transplant survival percentage of seedlings was more than 98%.[Conclusion] The domestication and transplanting of seedlings were carried out.This technical system can provide reference to the conservation and use of wild Lilium sulphureum resouces in Guizhou province.
