高钾停搏液对糖尿病大鼠离体心肌保护效果的研究

摘要 目的探讨STH-2(St. Thomas Hospital 2 cardioplegic solution)和HTK(Histidine-Tryptophan-Keto-glutarate Solution)对非糖尿病和糖尿病大鼠离体心肌保护后心脏功能的改变。方法健康雄性SD大鼠18只,随机分为3组(n=6,Control组,STH-2组,HTK组),链脲佐菌素诱导的糖尿病大鼠18只,亦随机分为3组(n=6,T-Control组,T-STH-2组,T-HTK组);用改良Langendorff装置测定心功能指数,留取冠脉流出液测定肌钙蛋白(TnI)含量。结果非糖尿病组心脏保存2h后心功能指数与Control组相比均有不同程度的下降,且除心率(HR)外各指标差异均有显著性(P<0.05);糖尿病组心脏保存4h后心功能指数与T-Control组相比亦有不同程度的下降,但均无统计学意义(P>0.05)。糖尿病各组心功能指数(除HR外)均较对应的非糖尿病各组要低(P<0.05),TnI含量则高(P<0.05);STH-2组TnI含量较Controll组和HTK组显著升高(P<0.01);T-STH-2组TnI含量较T-Control与T-HTK组显著升高(P<0.01)。结论糖尿病心肌基础心功能损伤严重,但对缺血再灌注耐受性好。 Objective To study the preservation effect of the STH-2 and HTK on the diabetic rat and nondiabetie rat. Methods 18 hearthy SD rats were randomly divided into 3 groups(n = 6, Group Controll, Group STH-2, Group HTK) , 18 diabetic rats induced by streptozotoein were also randomly divided into 3 groups( n = 6, Group T-Controll, Group T-STH-2, Group T-HTK). Cardiac function index was determined by Langendorff apparatus. TnI was measured by collected coronary artery. Results Cardiac function index decreased after preserved for 4 hours in diabetic groups compared with Group control( Table 1-1 ). And every cardiac function index except HR had significant difference (P 〈 0.05 ). Cardiac function index also decreased after preserved for 4 hours in nondiabetie groups compared with Group T-Control ( Table 1-1 ). But no significant difference existed ( P 〉 0.05 ). The diabetic groups had low cardiac function index and high TnI content compared with nondiabetie groups ( P 〈 0.05 ). Group STH-2 had high TnI content compared with Group Controll and Group HTK( P 〈0.01). Group T-STH-2 also had high TnI content compared with Group T-Controll and Group T-HTK( P 〈 0.01 ). Conclusion Diabetic myoeardium has lower basic eadiae function index but it has better toleration with isehemia/perfusion.
