摘要
目的:研究注射用苦碟子(Ixeris sonchifolia extracts for injection,KDZ)对心肌缺血再灌注损伤大鼠心肌梗死面积、心肌酶和血黏度的影响。方法:大鼠以KDZ(2.5、5.0、10g生药/kg·d,ip),或0.9%NaCl治疗2天后,结扎大鼠冠状动脉左前降支60min,再灌注120min造成心肌缺血再灌注模型,然后再立即给药一次。计算心肌梗死面积(MIS),测定血清肌酸磷酸激酶(CPK)、乳酸脱氢酶(LDH)、天门冬氨酸氨基转移酶(AST)活性及低、中、高切变率下全血黏度和血浆黏度。结果:与再灌注模型组比较,KDZ中、大剂量能明显缩小MIS,显著降低血清CPK、LDH及AST活性,小剂量也能降低血清LDH活性;KDZ大剂量使全血低、中、高切黏度及血浆黏度明显降低,中剂量也能降低全血低切黏度。结论:KDZ可缩小心肌缺血再灌注损伤大鼠的MIS,降低血清CPK、LDH、AST活性及血液黏度,发挥保护作用。
Objective:To study the protective effect of Ixeris sonchifolia extracts (KDZ) for injection on myocardial ischemia reperfusion injury in rats. Methods:Mter 2 days treatment with KDZ (2.5,5.0 and 10g crude drug/kg · d,ip ) or 0.9% NaCl in rats,the myocardial ischemiareperfusion model was induced by occulusion of left anterior descending coronary for 60min and reperfusion for 120min, followed by giving KDZ or 0.9% NaCl once again. Then the myocardial infarct size(MIS) was detected; the activities of creatine phosphokinase (CPK), aspartate aminotransferase (AST) , and lactate dehydrogenase (LDH) in blood serum were examined, and the high,medial, and low shearing blood viscosities and plasma viscosity were determined. Results:compared with that of the model control, the MIS and the activities of CPK, AST, LDH were significantly reduced ( P 〈 0.05 or P 〈 0.01 ), and viscosities of blood and plasma were declined ( P 〈 0.05 or P 〈 0.01 ) in rats treated by KDZ. Conclusion:KDZ has protective effect on myocardial ischemia reperfusion injury in rats.
