温阳抗寒合剂对哮喘模型大鼠sVCAM-1及IL-4与IL-5的影响 被引量:12

摘要 目的:探讨中药温阳抗寒合剂治疗哮喘的作用机制。方法:50只雄性SD大鼠,随机分为正常组、模型组、中药小剂量组、中药大剂量组和西药对照组。除正常组外,其余各组大鼠造模,并予相应药物干预。实验结束后收集标本,运用双抗体夹心酶联免疫法检测各组大鼠血浆中可溶性血管细胞黏附分子-1(sVCAM-1),以及血浆与肺泡灌洗液中白细胞介素(IL-4、IL-5)的含量。结果:与模型组比较,中药大剂量组与西药对照组均可显著降低sVCAM-1与IL-4、IL-5的含量(P<0.01),两药物组间比较无差异(P>0.05);而中药小剂量组改变不明显(P>0.05)。结论:中药温阳抗寒合剂可通过下调sVCAM-1、IL-4、IL-5的含量,减轻气道炎症,从而发挥治疗哮喘的作用,并有剂量依赖性。 Objective: To elucidate the mechanism of "Wenyang Kanghan Mixture" in treating bronchial asthma. Methods: Fifty rats were randomly divided into normal grouop, model group, high-dose Mixture group, and low-dose Mixture gruop, and Dexamethasone group. The contents of serum and bronchoalveolar lavage fluid interleukin-4 (IL-4), interleukin-5 (IL-5),soluble vascular cellular adhension molecule(sVCAM-1) were examined by means of ELISA. Results: The contents of serum sVCAM-1, IL-4 and IL-5 were lower in both high-dose Mixture group and Dexamethasone group(P〈0.01), and there was no difference between the two groups(P〉0.05). There were no significant changes in contents of sVCAM-1, IL-4 and IL-5 in low-dose Mixture group. Conclusion: "Wenyang Kanghan Mixture" may treat asthma by down-regulating the contents of sVCAM-1, IL-4 and IL-5 in a dose-dependent manner.
