摘要
目的探讨低浓度紫杉醇对小鼠血管功能及心肌的影响及局部病理解剖变化。方法36只小鼠随机分为2组。两组小鼠每日均饲喂抗血小板凝聚药物(阿司匹林肠溶片和氯吡格雷),实验组腹腔内注射紫杉醇(100μg/50g),对照组注射同量生理盐水,3个月后,分别检测血清心肌酶学变化,同时取心脏、血管行病理检查,检测心血管内皮形态学差异及冠状动脉内皮细胞凋亡发生率。结果紫杉醇实验组心肌酶谱表达水平(CK,CK-MB,AST,LDH)显著高于对照组(P<0.01),其中CK-MB高达12101.67±863.33,CRP均过低,差异无统计学意义(P>0.05);实验组冠状动脉内皮细胞凋亡发生率高于对照组,实验组见血栓形成(11.0%)高于对照组(0%)。结论低浓度紫杉醇长期作用可以显著提高小鼠某些心肌酶指标的水平,造成心肌损伤并促进冠状动脉内皮细胞凋亡,可能与晚期支架内血栓形成有关。
Objective To evaluate the effects of paclitaxinel on myocardial zymograms and endothelial morphology. Methods 36 mice were randomly involved in both experimental and control groups with daily anticoagulant feeding by water, then the differences of myocardial zymograms and endothelial morphology were compared between the two groups. Results The expression of zymograms (CK,CK-MB,AST,LDH) in experiment group were much higher than those of control (P〈0.01). Only the difference of CRP were not statistically significant (P〉0.05) between the two groups. The endothelial morphology apoptosis in experimental groups are higher than control groups. Conclusion Exposure to paclitaxinel in a long time would significantly increase the expression of myocardial zymograms ang increase endothelial morphology apoptosis, which play a role in the stent-thrombosis.
