骨形态发生蛋白和碱性成纤维细胞生长因子激发髓核细胞成骨潜能的实验研究

Experimental Study on the Provocation of Osteogenic Potential of Nucleus Pulposus Cells by Bone Morphogenetic Protein and Basic Fibroblast Growth Factor

摘要 【目的】研究联合使用重组人骨形态发生蛋白(rh-BMP2)和碱性成纤维细胞生长因子(bFGF)对椎间盘髓核细胞成骨潜能的激发作用。【方法】向体外培养的髓核细胞中分别及联合加入rh-BMP2和bFGF观察髓核细胞的形态学变化、细胞生长曲线变化、钙盐沉积情况、I型胶原分泌情况、骨钙素活性变化。【结果】联合使用rh-BMP2和bFGF能够明显促进髓核细胞增殖,由长梭形向多角形变化,细胞体积增大,细胞颗粒增多,增加I型胶原分泌,提高钙盐沉积程度,提高骨钙素的表达水平。【结论】联用rh-BMP2和bFGF能够诱导髓核细胞向成骨细胞方向分化,分泌钙盐并形成矿化结节。 [Objective] To evaluate the provocative effect of combined recombinant human bone morphogenetic protein-2 (rh-BMP2) and basic fibroblast growth factor (bFGF) on osteogenic potential of nucleus pulposus (NP) cells. [Methods] Simple or combined rh BMP2 and bFGF were added to NP cells cultured in vitro. Morphological alterations, cell growth curve, calcification of the complexes, secretion of type Ⅰ and Ⅱcollagen and activity of osteocalcin in NP cells were observed. [Results] The combined use of rh-BMP2 and bFGF significantly promoted the proliferation of NP cells, the alteration of cells from fusiform to polygon, the aug mentation of cell volume, and secretion of type I collagen, the deposition of calcium salt and the expression of osteocalcin. [Conclusion] The rh BMP2 and bFGF could induce the NP cells" differentiation to osteoblasts, and promote the formation of calcium salt and mineralized nodes.
