Ang-2、VEGF及MVD在胃癌中的表达及其临床意义 被引量:12

摘要 目的探讨血管生成素-2(Ang-2)、血管内皮生长因子(VEGF)在胃癌组织中表达及其与微血管密度(MVD)和临床病理特征的关系。方法应用免疫组化两步法检测51例胃癌组织中Ang-2、VEGF和CD34的表达,并以24例正常胃黏膜标本作对照。对CD34阳性血管进行MVD计数,并结合临床资料进行统计学分析。结果胃癌组织中的Ang-2及VEGF阳性表达率分别为60.8%、53%,MVD为31.9±11.2;正常胃黏膜组织Ang-2及VEGF阳性表达率分别为12.5%、4.17%,MVD为13.1±4.3,胃癌组织中Ang-2、VEGF表达率及MVD均明显高于正常胃黏膜组织(均P<0.01)。Ang-2表达与胃癌的大小、有无淋巴结转移、浸润深度、TNM分期有关(均P<0.01),而与患者年龄、性别、胃癌的分化程度无关。在胃癌组织中Ang-2的表达与VEGF、MVD呈正相关(分别为r=0.846,P<0.01;r=0.71,P<0.01)。VEGF表达与胃癌的浸润深度、分化程度、胃癌的大小、有无淋巴结转移、TNM分期有关(P<0.05或P<0.01),胃癌组织中VEGF的表达与MVD呈正相关(r=0.858,P<0.01)。MVD与胃癌的浸润深度、有无淋巴结转移、TNM分期有关(均P<0.01)。结论在胃癌组织中,Ang-2、VEGF的过度表达可能在肿瘤的血管生成和进展过程中起着重要作用。 Objective To investigate the expression of Angiopoietin-2 ( Ang-2 ), vascular endothelial growth factor(VEGF) in gastric carcinoma and to explore their relationship with microvessel density(MVD) and clinicopathological features. Methods The expressions of Ang-2 ,VEGF were studied by immunohistochemistry in 51 gastric carcinoma specimens and 24 normal tissues as control. The microvessel density(MVD) was assayed by CD34 positive blood vessels counting. Results The positive rates of Ang-2, VEGF were 60.8% , 53% respectively,and MVD was 31.9 ± 11.2 in gastric carcinoma. However, in normal gastric tissues, they were 12.5% ,4. 17% and 13.1±4.3 respectively. Expressions of Ang-2, VEGF proteins were significantly higher in gastric carcinoma than those in normal tissue( all P 〈0.01 ). The expression of Ang-2 was related to tumor size, penetrating depth,lymph node metastasis and TNM stage( all P 〈 0.01 ). In gastric carcinoma, Ang- 2 expression had a positive correlation with VEGF and MVD ( r = 0. 846, P 〈 0.01 and r = 0.71, P 〈 0.01 respectively). The expression of VEGF was correlated with penetrating depth, differentiation, tumor size, lymph node metastasis and TNM stage (P 〈 0.05 or P 〈 0.01 ). There was a positive correlation between VEGF expression and MVD in gastric carcinoma( r = 0. 858, P 〈 0.01 ). MVD was related to penetrating depth, lymph node metastasis and TNM stage (P 〈0.01). Conclusion Over-expressions of Ang-2, VEGF proteins mayplay an important role in angiogenesis and progression of gastric carcinoma.
