白细胞介素-1受体拮抗剂对大鼠退变椎间盘治疗作用的研究 被引量:2

摘要 目的探讨白细胞介素-1受体拮抗剂(IL-1Ra)对已开始退变椎间盘细胞基质代谢的影响。方法36只大鼠采用去双上肢及折尾法建立大鼠退变椎间盘模型,随机分为实验组和对照组,每组18只。实验组从造模3个月后给予腹腔注射IL-1Ra(50 ng.kg-1),隔日1次,对照组不给予任何处理。造模后4、5、6个月每组分别处死6只大鼠,完整取出L4-5椎间盘后进行固定、切片,HE染色观察椎间盘形态学变化,免疫组化染色法测定Ⅱ型胶原的含量;取出L5-6椎间盘,使用间苯三酚法测量蛋白多糖含量。结果实验组大鼠椎间盘Ⅱ型胶原灰度值低于对照组,蛋白多糖含量高于对照组,差异有统计学意义(P<0.05)。结论IL-1Ra可以延缓已退变的大鼠椎间盘继续退变,本研究为IL-1Ra应用于临床治疗椎间盘退变提供了理论依据。 Objective To explore the effects of the interleukin-1 receptor antagonist protein (IL-1Ra) on the degenerative intervertebral disc of the bipedal rat. Methods Degenerative model of bipedal rat was stablished. Thirty-six rats were randomly divided into experimental group and control group, each group had 18 rats. The rats in experimental group were intraperitoneally injected with IL-1Ra (50 ng·kg^-l) after three months of operation and repeated on alternative days until they were killed, while the rats in control group accepted no treatment. Six rats were killed from every group at four, five, six months post-operation respectively. The tissue of L4/5, L5/6 discs were fixed and sliced. HE Staining was used to investigat the change of morphologic. Type Ⅱ collagen was detected by immunohistochemistry staining, and the content of proteoglycan was measured by phloroglucin. Results The gray scales of type Ⅱ collagen in experimental group was lower than those in control group (P 〈 0.05 ). The content of proteoglycan was higher in experimtal group than in contron group (P 〈 0.05). Conclusion IL-1Ra protein can inhibit and reverse the degradative intervertebral disc of the bipedal rat, and it adds evidence that IL-1Ra can be used in clinic to treat degenerative intervertebral disc.
