摘要
目的探讨重组人粒细胞集落刺激因子(rhG-CSF)是否能改善局灶性脑缺血糖尿病大鼠的神经功能,并观察对神经细胞凋亡及胰岛素样生长因子-1(IGF-1)蛋白表达的影响。方法线栓法复制糖尿病大鼠大脑中动脉栓塞模型,rhG-CSF干预组连续皮下注射rhG-CSF,50μg/(kg·d),进行神经功能缺损评分,采用TUNEL染色计数脑缺血周边区神经细胞凋亡数,免疫组织化学方法检测IGF-1的蛋白表达。结果rhG-CSF干预组神经功能缺损评分明显低于对照组(P<0.01);rhG-CSF干预组缺血周边区的TUNEL阳性细胞数均明显少于对照组(P<0.01);IGF-1蛋白免疫阳性细胞明显多于对照组(P<0.01)。结论rhG-CSF可增加糖尿病脑缺血之后缺血周边区IGF-1蛋白的表达,拮抗神经细胞凋亡,改善神经功能。
[Objective] To investigate whether G-CSF could improve neurological deficits, inhibit neuronal cell apoptosis and increase the expression of IGF-1 after cerebral ischemia in diabetic rats. [Methods] Wistar diabetic rats (n =36) were subjected to middle cerebral artery occlusion and randomly divided into two groups: control group (n =18), G-CSF group (n =18). G-CSF group received subcutaneous injection of rhG-CSF 50 μg/kg·d after cerebral ischemia, neurological severity scores. TUNEL staining was used to count the number of apoptosis neurons in peri-ischemic area and immunohistochemical assessments was performed to examine the protein expression of IGF-1. [Results] The neurological score significantly increased in G-CSF groups compared with the control group(P 〈0.01). The expression of IGF-1 significantly increased, apoptosis cells significantly decreased in G-CSF group compared with control group(P 〈0.01). [Conclusions] RhG-CSF can improve neurological function through inhibition of neuronal cell apoptosis and increasing the expression of IGF-1 after cerebral ischemia in diabetic rats.
