摘要
目的探讨CD147蛋白、MMP-9 mRNA在子宫颈鳞状细胞癌中的表达及其与侵袭、转移的关系。方法采用免疫组化MaxVision法、RT-PCR技术检测10例慢性子宫颈炎组织(对照组)和40例子宫颈鳞状细胞癌患者的外科手术切除标本组织中CD147蛋白、MMP-9 mRNA的表达,应用图像分析软件分别对CD147和MMP-9 mRNA的表达进行平均光密度和灰度分析。结果CD147蛋白和MMP-9 mRNA的表达结果显示:子宫颈鳞状细胞癌组高于对照组慢性子宫颈炎组织(P值均<0.01);有淋巴转移组高于无转移组(P<0.01、P<0.05);CD147蛋白在宫颈鳞状细胞癌低分化组表达与中分化组表达差异无显著性(P>0.05),但MMP-9 mRNA表达为低分化组高于中分化组(P<0.05)。结论CD147蛋白和MMP-9 mRNA在子宫颈鳞状细胞癌中呈高表达,它们之间的表达具有正相关性,二者可能共同参与了子宫颈鳞状细胞癌的侵袭和转移过程。
Purpose To investigate the expression of CD147 and MMPo9 mRNA in cervical squamous cell carcinoma tissues and further to explore the correlation between their expression and the invasion and metastasis of the cancer. Methods Immunohistochemical MaxVision method and RT-PCR were used to detect the expression of CD147 and MMP-9 mRNA in both 10 cases of chronic cervical inflammation and 40 cases of cervical squamous cell carcinoma. The quantitative analysis of mean density of the expression of CD147 and the grey of MMP-9 mRNA were conducted, respectively, with the image analytic software, Results The levels of the expression of CD147 and MMP-9 mRNA were both obviously higher in cervical squamous cell carcinoma tissues than that in chronic cervical inflam- mation tissues( P 〈0. 01 ) ; their expression level was also clearly higher in the tissues of cervical squamous cell carcinoma with lymph node metastasis than that in those without lymph nodes metastasis ( P 〈 0. 01, P 〈 0. 05 ). There was no difference in the expression level of CD147 between low differentiation group in cervical carcinoma and moderate differentiation group ( P 〉 0. 05 ) , but the expression of MMP-9 mRNA in low differentiation group is higher than moderate differentiation group(P 〈0. 05). Conclusions There is a high level of expression of CD147 and MMP-9 mRNA in cervical squamous cell carcinoma, in which both CD147 and MMP-9 are positively related to jointly involve the invasion and metastasis of cervical squamous cell carcinoma.
