摘要
为了观察大鼠模拟失重后侧脑室下区(subventricular zone,SVZ)OX42阳性细胞的表达变化,本研究采用雄性SD大鼠尾部悬吊法建立模拟失重模型,按体重配对原则随机将20只大鼠分为4组:即模拟失重2周组(TS2W),模拟失重4周组(TS4W)和相应的正常对照(CON2W和CON4W)2组。用特异性标记小胶质细胞的OX42抗体行免疫组织化学ABC法染色,观察大鼠模拟失重后OX42阳性细胞在SVZ的形态和数量变化。结果显示:TS2W和TS4W组侧脑室下区OX42阳性小胶质细胞数显著增多,并呈显著活化状态,表现为阿米巴样小胶质细胞。本研究结果提示小胶质细胞可能参与失重的病理生理过程。
To investigate the expressional change of OX42-positive cells in subventricular zone (SVZ) of adult rats after been exposed to simulated microgravity, tail-suspension model was used to simulate microgravity in present study. Experiments were conducted on 20 male SD rats which were randomly divided into 4 groups: simulated microgravity for 2 weeks (TS2W) and 4 weeks (TS4W), control of 2 weeks (CON2W) and 4 weeks (CON4W). OX42 immunohistochemical (ABC) method was used to observe the morphological and number change of OX42-pasitive cells in SVZ in different groups. The results showed that the number of OX42-positive cells in SVZ of TS2W and TS4W increased significantly compared to control groups,and OX42-positive cells of TS2W and TS4W showed large ameboid morphology that was previously known as been in activated state. These results indicate that microglia maybe participate in the pathophysiological process of simulated microgravity exposing.
