绵羊肺腺瘤病毒囊膜基因表面蛋白的表达及其单克隆抗体的制备 被引量:3

摘要 在E.coli BL21中诱导表达含有绵羊肺瘤病毒囊膜表面蛋白(SU)重组质粒pGEX-4T-1-SU,表达的GST融合蛋白经纯化后用佐剂乳化,作为抗原免疫BALB/c小鼠,并进行细胞融合、克隆化、ELISA法初步筛选抗SU单抗。最终获得了3株抗SU的单抗,为深入探讨JSRV的分子生物学特性及研制诊断试剂盒打下了基础。 Monoclonal antibodies against Jaagsiekte sheep retrovirus were prepared using recombinant env protein expressed from plasmid pGEX-4T-1-SU in E.coli BL21. BALB/c mice were immunized and three McAb clones against SU domain of the env protein were developed by standard hybridoma technique and identified by ELISA. These McAbs provide potential useful tools for diagnosis of JSRV and for the further study of JSRV at molecular level.
