肿瘤坏死因子相关凋亡诱导配体对人宫颈癌HeLa细胞的凋亡作用 被引量:4

摘要 目的观察不同浓度肿瘤坏死因子相关凋亡诱导配体(TRAIL)对宫颈癌HeLa细胞的凋亡作用。方法2006年于重庆医科大学,采用不同浓度的TRAIL作用于HeLa细胞,采用MTT(噻唑蓝)法检测细胞存活分数;AO/EB(丫碇橙/嗅乙啶)双重荧光染色观察凋亡细胞结构;流式细胞仪检测细胞周期及细胞凋亡率。结果TRAIL作用于HeLa细胞24h后,TRAIL对细胞生长抑制率及细胞凋亡率具有量效关系;AO/EB双重荧光染色可见到早期、晚期凋亡细胞;流式细胞仪检测有亚二倍峰出现;细胞周期发生改变。结论TRAIL对HeLa细胞生长有抑制作用,并可诱导其凋亡。 Objective To observe the effects of different concentration of TRAIL on cen, ical cancer HeLa cells. Methods With different concentration of TRAIL role in HeLa cells, the survival fraction of HeLa cells was measured by MTT assay in Chongqing Medical University in 2006. Structures of apoptosis cells were observed by AO/EB fluorescence microscopy. Cell cycle of HeLa cells and apoptosis rates were determined by FACS. Results After treating Hela cells for 24 h by TRAIL, a good dose - effect relationship was demonstrated by the role of TRAIL in the rate of cell growth inhibition and apoptosis ; early and late apoptosis cell can be seen by AO/EB stainning and the hypodiploid peaks were found by FACS checking. The cell cycles were changed. Conclusion The proliferation of HeLa cells can be inhibited and the apoptosis can be induced by TRAIL.
