黄芪愈伤组织中黄芪多糖含量影响因素的研究 被引量:3

摘要 目的:研究培养基及成分和光照对黄芪愈伤组织中多糖类化合物形成的影响。方法:使用不同培养基并改变其中碳源、氮源、磷酸盐和钙盐的浓度,在黑暗和光照条件下进行黄芪愈伤组织培养,分光光度法检测愈伤组织中多糖类化合物的含量。结果:黄芪多糖积累的最佳条件是(24±1)℃光照培养,培养基为MS基本培养基(附加0.5 mg/L 2,4-D和1.0 mg/L 6-BA),其中含有80 g/L蔗糖、60 mmol/L KNO3、1.50 mmol/L KH2PO4和1.496 mmol/L CaCl2。结论:蔗糖作为碳源最有利于黄芪愈伤组织生物量积累和多糖类化合物的生物合成,硝态氮和光照有利于黄芪多糖类化合物的合成。 Objective: To study the effects of the culture com ponents and illumination on the accumulation of polysaccharide compounds. Method: By adjusting the concentration of carbon, nitrogen, phosphate and calcium in the culture solution, and kept the Astragalus callus under dark or illumination, then detected the content of polysaccharide compounds by spectrophotometry. Result: The optimum condition of Astragalus polysaccharides accumulation was (24±1)℃ in the light and the basic medium was Murashige and Skoog (MS) (plus 0.5 mg/L 2,4-D and 1.0 mg/L 6-BA) with 80 g/L sucrose, 60 mmol/L KNO3, 1.50 mmol/L KH2PO4 and 1.496 mmol/L CaCI2. Conclusion: Sucrose, source of carbon facilitated to the accumulation of biomass and synthesis of polysaccharide compounds. And both nitrate and illumination favored for the synthesis of polysaccharide compounds.
