摘要
17997为由我国云南省土壤中分离的一株链霉菌所产生的广谱抗病毒抗生素。在淋巴细胞培养内,17997对人免疫缺陷病毒I型(HIV-1)不同株有抑制活性,IC50为0.018~0.15μM,与叠氮胸苷(AZT)耐药株无交叉耐药,且与AZT有协同作用。在猴免疫缺陷病毒(SIV)急性感染的恒河猴模型内,腹腔给药及皮下给药均能降低血浆内病毒滴度,缩短病毒血症时间,以腹腔给药效果较皮下给药佳且有量效关系。结果说明17997在实验条件下有抗SIV感染作用。
is a broad spectrum antiviral antibiotic produced by a strain of streptomyces isolated from a soil sample collected in Yunnan Province of China.17997 displayed inhibitory effects on deferent strains of human immunodefficiency virus type 1(HIV-1)in deferent cell lines with IC 50 of 0.018-0.15μM.AZT resistant strain of HIV1 is still sensitive to 17997 without cross resistance.In cell culture both 17997 and AZT are synergistic against HIV1.In vivo experiment rhesus monkeys were infected by Simian Immunodeficiency Virus(SIVmac 251)and treatment by 17997 via ip or sc once a day for 22 days.The results showed that 17997 treatment could reduce the serum virus titers and shorten the period of viruemia.Theeffcacy of 17997 treatment by ip rout was better than did by sc rout.It also showed dose-efficacy relationship. This indicated that 17997 could inhibit SIV replication in the experiment conditions. \
