摘要
目的建立HPLC-MS/MS法测定人全血中唑吡坦的方法。方法采用HPLC-MS/MS法测定人全血中唑吡坦的浓度,色谱柱:XTerra RP18(2.1×100mm,3.5μm);流动相:甲醇:10m mol/L醋酸铵水溶液(用乙酸调pH=6.0)40:60;流速:0.2ml/min。结果最低检测限为0.5ng/ml,血药浓度线性范围5.0~500ng/ml,高、中、低3种浓度的回收率分别为(103.95±2.96)%,(100.79±4.27)%,(96.06±7.7)%。
Objective To develop a HPLC-MS/MS method for the determination of zolpidem in human blood. Method The whole blood sample was extracted with a liquid-liquid extraction. The extracts were analyzed by HPLC-MS with a XTerra RP18(2. 1 × 100mm,3.5μm) column and a mobile phase of methanol : 10 mmol/L ammonium acetate (pH= 6.0). Results The limit of detection(LOD) was 0. 5ng/ml, the calibration curve was linear over the range of 5. 0-500ng/m|,the recovery of high, medium and low concentrations were (103.95±2. 96)%, (100. 79±4. 27)%, (96. 06±7. 7) % , respectively.
