强制性运动疗法对大鼠脑缺血再灌注后神经修复及Rho激酶表达的影响 被引量:10

Effects of Constraint-induced Movement Therapy on Neural Reparation and Rho Kinase Expression after Cerebral Ischemia-reperfusion in Rats

摘要 目的探讨脑卒中后强制性运动疗法(CIMT)对大鼠脑缺血再灌注后行为功能及大脑皮质缺血区Rho激酶表达的影响。方法健康雌性SD大鼠75只,随机分为强制性运动疗法组(CIMT组)25只,缺血再灌注组(I/R组)25只和假手术组(Sham组)25只,对前两组大鼠用线栓法建立大脑中动脉缺血再灌注模型。3组大鼠再随机分为缺血再灌注后7,14,21,28和35d时处死组。各组动物在处死前行神经功能缺损评分(Bederson评分),水迷宫试验和平衡木试验,并采用Western blot方法检测Rho激酶的表达。结果与I/R组比较,在脑缺血再灌注术后14d和21d,CIMT组的肢体运动平衡能力及记忆功能明显恢复;在缺血侧大脑皮质中Rho激酶的表达,CIMT组在14d和21d时的表达明显低于I/R组。结论强制性运动疗法可以改善脑缺血再灌注后的神经功能,其在分子水平上发挥作用可能和下调Rho激酶的水平有关。 Objective To study the effects of constraint-induced movement therapy (CIMT) on the grade of behavior in rats with cerebral ischemia-repeffusion and the expression of Rho kinase around ischemic facus. Methods 75 Healthy female Sprague-Dawley rats were randomly divided into 3 groups : constraint-induced movement therapy group (CIMT group, n = 25 ), ischemia-repeffusion group (I/R group, n = 25 ) and sham group(n = 25 ). The CIMT group and I/R group were modeled with cerebral ischemia-repeffusion by blocking middle cerebral artery with a nylon thread. The three groups then were divided into 7th, 14th,21st,28th,35th day execution groups after cerebral ischemiarepeffusion by randomized methods. Behavior scores,trapezoid balance bar experiment and Morris water maze experiment were all carried out to evaluate the recovery of neural function at 7th, 14th, 21st, 28th, 35th day postsurgery respectively. The expression of Rho kinase in the peri-ischemia cortex of the three groups were also evaluated at the same day by western blot. Results Compared with I/R group, the motor function,balancing function and mnemonic function of CIMT group were all improved in 14th and 21st days after cerebral ischemia-repeffusion and the expression of Rho kinase in C1MT group decreased more obviously than the I/R group in 14th and 21st days after cerebral ischemia-repeffusion. Conclusion The constraint-induced movement therapy will improve the behavior of rats with cerebral isckemia-repeffusion,and the mechanism may have relation with down-regulation of the expression of Rho kinase around the ischemic focus.
