高压氧对缺血再灌注小鼠脑组织中细胞因子IL-1β、IL-8及血脑屏障通透性的影响 被引量:8

摘要 目的探讨高压氧(HBO)对缺血再灌注小鼠脑组织中细胞因子IL-1β、IL-8蛋白含量和mRNA的表达及血脑屏障通透性的影响。方法复制清醒小鼠脑缺血再灌注模型,于再灌注期间经0.25MPa(ATA)HBO治疗5次,采用ABC-ELISA、RT-PCR法检测脑组织中IL-1β、IL-8的含量、mRNA的表达及依文思蓝(EB)的含量。结果脑缺血再灌注组于再灌注72h脑组织中细胞因子IL-1β、IL-8含量及mRNA的表达较假手术组明显升高(P<0.01);并与脑组织EB含量渗出增多相一致。HBO+脑缺血再灌注组与脑缺血再灌注组相比细胞因子IL-1β、IL-8含量及mRNA的表达明显降低(P<0.05),并与脑组织EB含量渗出减少相一致。HBO组与假手术组相比细胞因子IL-1β、IL-8的含量及mRNA表达差异不显著(P>0.05)。结论HBO从细胞及基因水平可明显减少脑缺血再灌注损伤中促炎因子IL-1β、IL-8的含量及表达,从而对血脑屏障具有保护作用;而对正常脑组织中细胞因子IL-1β、IL-8的含量及表达影响很小。 Objective To investigate the effect of hyperbaric oxygen (HBO) on cytokines in the brain tissues and blood-brain barrier after cerebral ischemia-reperfusion in mice. Methods The conscious mice cerebral ischemia-reperfusion models were established,and 0.25 MPa (ATA) HBO was applied 5 times during the reperfusion period,and 2%Evans blue was injected into tail veins an hour before the animals were killed. The levels of IL-1β, IL-8 and the content of Evans blue of brain tissues were determined by ABC-ELISA, RT-PCR method and spectrophotometry. Results The levels of IL-1β,IL-8 and the content of Evans blue were obviously increased in ischemia-reperfusion group,compared with the sham group (P 〈 0.01 ). The levels of IL-1β, IL-8 and the content of Evans blue in HBO+ischemia-reperfusion group were obviously decreased compared with ischemia-reperfusion group (P 〈 0.05). The levels of cytokines and the content of Evans blue in brain in HBO group were similar to those of the sham surgery group (P 〉 0.05 ). Conclusion HBO (0.25 MPa 5 times) has significant effect on reducing the levels of IL-1β and IL-8 during cerebral ischemia-reperfusion,so it can reduce the permeability of blood-brain barrier,but has little effect on normal brain tissues.
