摘要
目的探讨雷帕霉素靶蛋白(mTOR)/S6蛋白激酶(p70S6K)信号通路在兔正畸牙移动过程中对牙周组织改建的作用。方法选用12只日本大耳白兔(体质量2.0kg左右),雌雄不限,建立正畸牙移动的动物模型,按照3,5,7,14d随机分组,每组3只,上颌右侧戴矫治器组为实验组,左侧未戴矫治器组为对照组,采用Western免疫印迹的方法,观察牙周组织中mTOR及p70S6K蛋白表达的变化。结果戴矫治器3d后牙周组织中mTOR和p70S6K表达增强,7d达高峰,14d后有所下降,与对照组比较有显著差异(P<0.01)。结论在矫治力作用下,兔牙周组织中mTOR及p70S6K表达均明显增强,导致牙周组织发生一系列变化。
Objective To explore the effect of mammalian target of rapamycin and p70 S6 Kinase on rabbit periodontal remodeling on orthodontic tooth movement. Methods Twelve rabbits (weight 2.0±0.2kg) were divided randomly into groups of 3,5,7 and 14 days with 3 rabbits in each group to establish rabbit models for the study. The appliances were located on the right maxillary in the test group and unfixed appliances on the left in the control group. The animals were killed at each time intervals (3,5,7 and 14 d) respectively. Western blot test was applied to evaluate the expression of mTOR and p70 S6K. Results The expression of mTOR and p70 S6K was observed changing dramatically; the expression of mTOR and p70 S6K in the periodontal tissues with orthodontic treatment was higher than that in the control, up to the peak on the 7th day,and then decreased,with statistical significance compared with the control group (P 〈 0.01). Conclusion mTOR and p70 S6K participate in the tissue remodeling during orthodontic tooth movement and may play a more important role in the process of periodontal remodeling.
